Pam-dependent Specificity
PAM-Dependent Specificity is a targeting constraint in CRISPR-Cas systems that refers to the requirement for a specific protospacer adjacent motif (PAM) sequence adjacent to the target DNA site, thereby distinguishing it from PAM-independent mechanisms by restricting cleavage activity to genomic loci flanked by a predetermined nucleotide signature.
