Single-agent vemurafenib leads to a rapid and sustained clinical response in severe multisystem LCH but does not eradicate the disease.Longitudinal assessment of <i>BRAF</i> V600E during treatment shows that clinical remission can occur despite significant amounts of mutated BRAF.
