In vivo and in vitro experiments demonstrated that rat liver cells are able to synthesize a double-labeled mannolipid from t3H]retinol and GDP-[14C]mannose.The doublelabeled mannolipid was purified by DEAE-cellulose, silicic acid, and thin-layer chromatography, and shown to contain retinol and mannose in a molar ratio in the range of 1: 1.The biosynthesis of the mannolipid, catalyzed by a liver membrane fraction, depended on ATP, Mn2+, and retinol, when the enzyme was prepared from vitamin A-deficient rats.
