Synthesis and Biological Activity of Largazole and Derivatives

Abstract

A modular synthesis of the marine natural product largazole and related synthetic analogues is described. Largazole was prepared in 19 % overall yield through a synthetic route with a longest linear sequence of nine steps. Activity tests showed the necessity of the thiobutenyl moiety for antiproliferative activity. Detailed facts of importance to specialist readers are published as ”Supporting Information”.
