An <i>S</i>‐Selective Lipase Was Created by Rational Redesign and the Enantioselectivity Increased with Temperature

Abstract

Aktiver mit größerer Tasche: Die Abbildung zeigt eine Überlagerung von Zwischenstufen, die beim durch Candida-antarctica-Lipase B (CALB) katalysierten Acyltransfer auf (S)-1-Phenylethanol auftreten. Die Wildtyp-CALB kann die Phenylgruppe (grau) nicht in der Stereospezifitätstasche unterbringen und alle katalytisch wesentlichen H-Brücken bilden. Die Trp 104 Ala-Mutation schafft den gelben Raum, das S-Enantiomer passt leicht hinein, und die Spezifitätskonstante steigt um den Faktor 130 000.
