Publisher Summary This chapter describes a selection of experiments chosen because they illustrate the scope both of spin-label investigations of membrane organization and of experimental techniques. The spectral parameters of choice for analysis of the paramagnetic resonance data of many spin label experiments are derived from theoretical considerations. Analysis of spectra for several of the experiments described in this chapter requires simulation of observed spectra by computer calculation. The approach to and assumptions of these calculations are presented as an introduction to the paramagnetic resonance spectra of spin labels. A more detailed discussion of basic theory for spin-label spectra has been also presented. General introductions to paramagnetic resonance theory may be found in books by Carrington and McLachlan and by Slichter.
