Abstract We report three individuals (two brothers and their father) with cerebral gigantism. All three had striking macrocephaly, large birth weight, gigantism, and variaable degrees of psychomotor retardation. Other findings included abnormal dermatoglyphics and an unusual metacarpophalangeal pattern profile in two of the three. To our knowledge, this is the first family with documented male to male transmission of cerebral gigantism.
