Detection of Metal-Bound Histidine in Ultraviolet Resonance Raman Spectra: Superoxide Dismutase

Ultraviolet resonance Raman (UVRR) spectroscopy reveals vibrational modes of metal-bound histidine side chains when the latter are equilibrated in D2O. These UVRR bands are seen clearly for Cu,Zn-superoxide dismutase, and their frequencies and intensities differ for the two metal ions. These modes offer a new selective probe for histidine ligands in metalloproteins.
