Abstract The synthesis of a number of 3‐(piperidyl‐(2)‐methyl‐) indoles and of some related compounds is described, together with that of an additional number of 3‐(piperidyl‐(N)‐methyl‐) indoles (cf.). An analysis of the relationship between structure and oxytocic activity (data of van Proosdij‐Hartzema and de Jongh ) shows that several structural and spatial details, decisive for the activity of the ergot alkaloids, have no such influence with the synthetic compounds, though the importance of the general pattern of the alkaloids is discernable. So the 3‐(piperidyl‐(2)‐methyl‐) indoles are found to be less active and less specific than the 3‐(piperidyl‐(N)‐methyl‐)‐indoles.
