SUMMARY The total gas output of slices of lactating rats' mammary glands metabolizing in bicarbonatesaline containing glucose and acetate is increased by the addition of insulin. From this response a straight-line assay has been developed. Using eight rats and twelve respirometers per rat, a six-point assay gave an estimate of relative potency consistent with that obtained by the manufacturers of the preparations used. A value of 0·18 was obtained for the index of precision (λ).
