Three polypeptides, derived from bovine encephalitogenic protein by proteolysis during its preparation have been isolated and their position of origin from the intact molecule has been determined. Fragments of the protein have also been prepared from both the undigested protein and the isolated polypeptides by chemical degradation using the bromine adduct of 2-(2-nitro-phenylsulphenyl)-3-methylindole or cyanogen bromide. Thus a set of polypeptides covering the following different regions of the encephalitogenic protein has been obtained: residue numbers, 1–116, 1–43, 1–20, 21–43, 44–170, 44–116, 90–170 and 117–170.
