The Respiratory Effects of Pentazocine

Abstract

SUMMARY The respiratory effects of pentazocine were compared with those of pethidine in 10 volunteers. Five of these produced C0 2 response curves at intervals of up to 21/2 hours after intravenous injection of the drug; the other five were subjected to continuous infusion of the drug while the end‐expiratory P CO 2 was maintained at 45 mm Hg. A dose of 15 mg/70 kg of pentazocine was found to produce respiratory depression of a degree similar to that of 50 mg/70 kg of pethidine in the first type of experiments, and likewise, the effect of 45 mg/70 kg of pentazocine corresponded well to that of 150 mg/kg 70 in the second type of experiments. ZUSAMMENFASSUNG Bei 10 freiwilligen Versuchspersonen wurden die Wirkungen von Penta‐zocin auf die Atmung mit denen von Pethidin verglichen. Bei 5 dieser Personen wurden CO 2 ‐Provokationskurven in Intervallen bis zu21/2 Stunden nach der intravenösen Injektion des Mittels hergestellt, bei den anderen 5 wurde das Mittel als Dauertropfinfusion verabreicht und dabei das endexspiratorische p CO 2 bei 45 mm Hg gehalten. Bei der ersten Versuchsreihe war eine Dosis von 15 mg pro 70 kg von Pentazocin imstande, eine Atemdepression zu er‐zeugen, die der Wirkung von 50 mg pro 70 kg von Pethidin vergleichbar war. Bei der zweiten Versuchsreihe lag Wirkungsgleichheit von 45 mg pro 70 kg Pentazocin mit 150 mg pro 70 kg von Pethidin vor.
