The Influence of Age on the Pharmacokinetics of Nitrendipine

The influence of age on the pharmacokinetics of nitrendipine has been studied by comparing plasma concentrations on day 1 and day 8 of a course of treatment with nitrendipine 20 mg daily in two groups. The young group contained eight healthy young volunteers — five male and three female — with a mean age 20 years and a mean weight 76 kg. This group came to the trials room for the two study days and rested supine for the first 8 h. The elderly group, two male and six female, were patients with mild hypertension, aged 67–83 (mean 77) years and with a mean weight of 73 kg. This group was admitted to the hospital for the whole study so that the effects of the drug could be monitored. The mean plasma concentration-time curves for the young and the old were very similar both on day 1 and day 8. The mean areas under the curve (± SD) on day 1 were 96.0 ± 66 ng. h ml−1 for the young and 97.8 ± 54 ng.h.ml−1 for the old. The corresponding figures on day 8 were 110.8 ± 86 and 143.2 ± 74. Age therefore does not seem to influence the disposition of nitrendipine.
