When IS1 is present in a particular DNA sequence, numerous DNA rearrangements occur in its vicinity at frequencies several orders of magnitude higher than when IS1 is absent (Reif and Saedler 1975, 1977; Cornelis and Saedler 1980). This paper reports on investigations of IS1-induced deletions, transpositions of a given IS1 to nearby sites in both orientations, and IS1-mediated inversions. In addition, an analysis of the regions within IS1 and in the target sites involved in these rearrangements is presented that leads to a model for the mechanism of IS1 integration.
