Orthosteric Coupling

Orthosteric Coupling is a mechanistic principle in allosteric protein engineering that denotes the direct energetic and structural interdependence between the orthosteric site and a distant functional site within a protein, where ligand binding or perturbation at the orthosteric site allosterically modulates the protein’s activity through defined conformational or dynamic pathways.
