Visualization and Replication era
The Visualization and Replication era (1935–1966) integrated visualization-driven cytogenetics with replication dynamics through polytene mapping, banding observations, and autoradiography. Barbara McClintock illuminated the link between chromosome structure and function in maize by showing how chromosomal rearrangements and transposable elements drive variegation and gene expression. Curt Stern advanced Drosophila cytogenetics with salivary-gland polytene chromosome mapping, demonstrating how visible bands and puffs reflect gene order and replication timing. Emil Heitz defined heterochromatin and chromosomal banding patterns, foreshadowing how structural features relate to replication domains, while Hermann Muller used radiation-induced aberrations to probe how chromosome structure constrains inheritance.
