23:T463,Bacteriophage therapy and application of phages for biocontrol necessitate acquisition of suitable phages. The exclusivity of phage-host relations and the risk of phage resistance instigate a need to rapidly isolate and characterize novel phages and continually build sizeable phage libraries. Current methods for phage isolation are both laborious and time consuming, suitable for the isolation of a limited number of phages. The high-throughput screening method for phages upscales and organizes enrichment of phages for fast isolation and identification of potentially hundreds of distinct phages against single hosts. This enables screening of hundreds of samples, in multiple simultaneous setups with varying parameters, increasing the likelihood of isolating multiple distinct phages specific for the given conditions. The efficiency of the method is emphasized by our screening of 200 environmental samples, resulting in the identification of an abundance of unique phage species virulent to Escherichia coli, Salmonella enterica, Enterococcus faecalis, and Pseudomonas aeruginosa.24:T48c,Urinary tract infections (UTIs) are a problem worldwide, affecting almost half a billion people each year. Increasing antibiotic resistance and limited therapeutic options have led to the exploration of alternative therapies for UTIs, including bacteriophage (phage) therapy. This systematic review aims at evaluating the efficacy of phage therapy in treating UTIs. We employed a comprehensive search strategy for any language, any animal, and any publication date. A total of 55 in vivo and clinical studies were included. Of the studies, 22% were published in a non-English language, 32.7% were before the year 1996, and the rest were after 2005. The results of this review suggest that phage therapy for UTIs can be effective; more than 72% of the included articles reported microbiological and clinical improvements. On the other hand, only 5 randomized controlled trials have been completed, and case reports and case series information were frequently incomplete for analysis. Overall, this comprehensive systematic review identifies preliminary evidence supporting the potential of phage therapy as a safe and viable option for the treatment of UTIs.25:T44f,Background: When subjected to phage infection, bacteria can rapidly become resistant by changes in the phage receptors at the bacterial surface. Phages thus require adaptive mechanisms to circumvent this type of resistance. Methods: LUZ7 phage with an altered host range were isolated and analysed for mutations and their effect. Results: We find that Pseudomonas virus LUZ7 has an unusually high number of mutants (0.01-0.1% of the population) that drive host range expansion. Interestingly, all tested mutants have a single D737Y mutation in the tail fiber. This mutation allows the phage to adsorb to P. aeruginosa strains that are not natively recognized by the wild-type phage. Conclusion: The high number and specificity of mutants suggests the presence of an uncharacterized mechanism that drives these mutations. This mechanism enables the phage to better evade host resistance at the surface level and expand its host range in general, a feature that could be valuable in phage therapeutic settings or for phage engineering.
