Abstract

Electrospinning provides exceptional flexibility in fabricating wound dressings. However, constructing 3D fibrous dressings presents a challenge. Herein, a multifunctional electrospinning fiber sponge (EFS) is developed by combining the humidity-controlled electrospinning with freeze-drying solidification for chronic wound healing. The moisture-induced phase separation triggered the spontaneous crimping and stacking of polylactic acid electrospun fibers into a unique fluffy structure, endowing EFS with excellent water absorption and blood-absorbent hemostatic properties. Subsequent freeze-drying further enhances the shape stability and compression elasticity of EFS. Compared to its electrospun membrane rival, drug (Curcumin and Ciprofloxacin hydrochloride)-loaded EFS (CC@EFS) exhibits superior antibacterial and antioxidant capabilities due to the effective release of therapeutic agents facilitated by its high porosity. In vivo experiments demonstrate that CC@EFS significantly accelerates infected wound healing by killing bacteria, alleviating inflammation, and promoting angiogenesis. Overall, this work paves an accessible way for engineering innovative 3D wound dressings for improved wound management.
