Infections caused by drug-resistant <i>Acinetobacter baumannii</i> have posed a serious threat to global public health. However, genetic manipulation methods, the primary way to study pathogenesis and drug-resistance mechanisms, remain time consuming and inefficient. Here, we provide a detailed protocol for genetic manipulation, including gene deletion, insertion, and point mutation in <i>A. baumannii</i> using the platform.
