Abstract

A gas chromatographic procedure for the estimation of N-hydroxyphentermine (2-methyl-2-hydroxyamino-1-phenylpropane) has been used to characterize the enzyme system present in liver microsome preparations that effects the N-hydroxylation of phentermine (2-methyl-2-hydroxyamino-1-phenylpropane) in vitro . When rabbit liver microsomes were used, the reaction was inhibited by SKF 525-A and by carbon monoxide. The rabbit microsomal preparation was unstable and decayed to 50% of original activity after 10 days of storage at -70°C. The relative order of N-hydroxylation activity in the three species examined was rabbit > guinea pig ≥ rat. Kinetic studies were also conducted.
