Transfer experiments in the reticulocyte ribosomal system using separate E. coli transfer RNAs for leucine indicated that one leucine position in the α-chain is different from all the other leucine positions in rabbit hemoglobin. In addition, the amino acid composition of the relevant peptides made us suspect that this leucine position might also be special in its structure (Weisblum, Gonano, von Ehrenstein, and Benzer, 1965).
