Simple method for electrophoretic analysis of serum lactic dehydrogenase.

Abstract A simple method is described for routine electrophoretic analysis of serum lactic dehydrogenase. The electrophoresis is conducted with an agar buffer solution, in an apparatus which combines electrode compartments and running surface into a single unit. A substrate-nitro-blue tetrazolium solution is employed for detection and quantitation of the separated isozymes. The apparatus can readily be constructed in any laboratory, and no special equipment is required. Twelve or more samples can be separated and detected in approximately one hour of working time. The sensitivity is sufficient for the detection of five isozymes in normal serum. The method is reproducible, and the optical density of the stained isozyme is nearly proportional to its concentration over a range of values.
