Two strains of herpes simplex virus (17, a typical type 1 strain, and HSG 52, a typical type 2 strain) are used in our Institute in Glasgow for the detailed study of the genetics and biochemistry of this large DNA virus. Each strain was initially purified by repeated single-plaque isolation, and this cloned virus was then used for isolating the mutants described in this paper.
