Phosphine, a fumigant to control insects, was found to inhibit the respiratory chain of rat liver mitochondria using succinate or pyruvate plus malate as substrate. The compound could not inhibit State 4 respiration of intact mitochondria, but it inhibited the respiration which was released by AMP, Ca2+ or DNP. When mitochondria were sonicated, the respiration was immediately inhibited by phosphine. It was considered that the changes in mitochondrial conformation from the controlled respiration to the released one allow the penetration of the inhibitor into the mitochondria.
