x-Chromosome Linked Inheritance of Elevated Thyroxine-Binding Globulin in Association with Goiter

Abstract

A new 4-generation family with high thyroxine-binding globulin, transmitted as an x-chromosome-linked codominant trait, has been studied. This kindred increases to 6 the number of high TBG families known, 3 of which provide clear evidence for x-chromosome linkage of the trait. The probability that this is a chance relationship is reduced to less than 1 in 5×108. A high incidence of simple or adenomatous goiter was also present in this family. Thirteen of 15 examined consanguineous family members with high TBG also had a goiter, whereas goiter was not found in 16 of 17 examined consanguineous family members with normal TBG. These results and the pedigrees strongly suggest that the goiter trait in this family was transmitted as an x-chromosome-linked character. The limited data available suggest that 2 separate genetic loci on the x-chromosome are involved.
