Abstract Shikimate dehydrogenase (E.C. 1.1.1.25) is found in plants but not in animals and is therefore an attractive target for a new herbicide. A series of 1,6‐dihydroxy‐2‐oxo iso nicotinic acid derivatives was prepared and shown to inhibit the enzyme reversibly. Weak active‐site directed irreversible inhibitors based on these compounds were also synthesised but none was herbicidal.
