The in vitro Product of SV40 DNA Transcription and Its Specific Hybridization with DNA of SV40-Transformed Cells

Transcription of SV40 and polyoma DNA in vitro by E. coli RNA polymerase yields an RNA product of high specific radioactivity. This product was successfully employed previously to detect minute quantities of virus DNA in virus-transformed cells (Westphal and Dulbecco, 1968; Sambrook et al., 1968). Little was known, however, about the actual nature of the in vitro reaction.
