Abstract

volume of distribution varies proportionally to lean body mass index [2,3], and clearance can be enhanced in paediatric patients [4 -6], resulting in important PK variability. All these changes can lead to decreased drug exposure and to the need for increased drug dose. Therefore, the dose ratio per kg is usually higher in CF patients, but this is partially contra-balanced by the relatively low body weight of CF patients (mean: 45 kg) when expressed as total dose per day.
