Abstract

Summary Attenuation of chicken anaemia agent (CAA) by serial passages in cell culture Chicken anaemia agent (CAA) was attenuated by serial passages in cultures of the MDCC‐MSB 1 cell line. Infectivity of CAA was assayed in cell cultures (TCID 50 ) and in day‐old chickens (chicken‐ID 50 ). Pathogenicity of CAA was significantly decreased as early as after 12 passages in cell culture, and it further decreased but was not completely lost during an additional 100 passages. Titres of virulent CAA propagated in chickens were in the mean by 1. 4 log lower in chickens than in cell culture. Differences between TCID 50 and chicken‐ID 50 of the 12th and 13th TC passage of CAA varied from 4.0 to 4.3 log and increased to more than 6.0 log after 100 to 112 passages. The chicken‐ID 50 of the 12th TC passage material of CAA as indicated by the incidence of infectious anaemia was increased by 1.4 log if birds were dually inoculated with CAA and Marek's disease virus. No differences could be detected between Gifu‐1 and Cux‐1 strains of CAA. Adaptation of CAA to growth in cell culture was not improved by serial passages. The possible use of cell culture‐passaged CAA as a vaccine for breeder flocks is discussed.
