By using complete model spectra, rather than just individual resonances, maximum information and uniqueness are incorporated into the analysis. A constrained regularization method accounts for differences in phase, baseline, and lineshapes between the in vitro and in vivo spectra, and estimates the metabolite concentrations and their uncertainties. LCModel is fully automatic in that the only input is the time-domain in vivo data. The lack of subjective interaction should help the exchange and comparison of results. More than 3000 human brain STEAM spectra from patients and healthy volunteers have been analyzed with LCModel. N-acetylaspartate, cholines, creatines, myo-inositol, and glutamate can be reliably determined, and abnormal levels of these or elevated levels of lactate, alanine, scyllo-inositol, glutamine, or glucose clearly indicate numerous pathologies. A computer program will be available.1e:T414,GALACTITOL has recently been isolated from the urine¹and brain²of galactosemia patients. Subsequently the conversion of orally administered galactose- 1-C¹⁴into galactitol-1-C¹⁴by a 17-year-old male galactosemia patient was demonstrated.³In order to account for the relatively slow excretion of administered radioactivity as galactitol-C¹⁴it was suggested that there had either been deposition of galactitol in tissues of the body, or that the administered galactose-C¹⁴had been incorporated into galactose-containing compounds. This hypothesis was further supported by the finding of galactitol in a variety of tissues in a rat fed large amounts of galactose.⁴We now wish to report the galactitol, glucose, free, and phosphatide-boundmyo-inositol content of body tissue from 17 areas of an infant presumed to be galactosemic.

Material and Methods

The patient studied was a 23-day-old male infant who was jaundiced on the third day of life.1f:T495,The presence of acyclic polyols in mammalian tissue was formerly considered a rare finding confined to specialized tissue (I). Van Heyningen reported the occurrence of significant quantities of galactitol in the lens of galactose-fed rats, although only minor amount,s of the hexitol were located in the heart and leg muscle (2). Other tissues which were examined, blood, liver, spleen, lung, and kidney, contained no detectable amounts of galactitol. Recently, however, galactitol was isolated from the urine of two’ male, galactosemia patients (3). By utilizing the convenient and sensitive technique of gas-liquid chromatography (4, 5) for the analysis of the urine and lens of rats fed a 35% galactose-containing diet, we have confirmed the ability of normal rats to convert significant amounts of galactose to galactitol.2 In addition, however, these studies led to the detection of large amounts of free galactitol in brain and kidney tissue. It therefore became of interest to examine organs of galactosemia subjects for possible deposition of galactitol. This report describes the analysis and isolation of galactitol from the brains of two galactosemia patients.
