The photophysical properties of the lowest excited triplet states of pyrido[3,4‐c]psoralen (PyPs) and 7‐methylpyrido[3,4‐c]psoralen (MePyPs) have been investigated by laser flash photolysis, including energy transfer from these triplets to oxygen‐generating singlet oxygen. A parallel study of the photosensitization of yeast in vivo by these compounds in the presence and absence of oxygen is also reported. The low triplet and hence singlet oxygen yields, reflected in the lack of an oxygen effect in yeast, suggest that photoadditions to DNA are likely to be the main source of the photosensitized lethal effects induced by these pyridopsoralens in vivo.
