Distribution of Barley Stripe Mosaic Virus Protein in Infected Wheat Root and Shoot Tips

SUMMARY Post-sectioning immunogold—silver staining (IGSS) specifically detected barley stripe mosaic virus (BSMV) protein in Lowicryl- or Araldite-embedded plant tissues at the light microscope level. Electron microscopy of adjacent sections showed that BSMV particles were present where the IGSS reaction was positive. BSMV protein was localized in or near the apical meristems of root and shoot tips of systemically infected wheat. Infected cells were distributed in a mosaic pattern.
