AN AUTOMATED, RAPID, AND QUANTITATIVE MICROASSAY FOR HISTOCOMPATIBILITY TESTING

Abstract

The lymphocytotoxicity microassay of histocompatibility testing was quantitated by the use of a new instrument, the Cytograf 6301. Reconstitution experiments and comparison with microscopic observations by technicians confirmed the sensitivity, accuracy, and reproducibility of the instrument. Measurements of HL-A antigenicity assayed on a microscale by the instrument were qualitatively similar to the standard microscopic microdroplet method. Moreover, the instrument allowed rapid and automated quantitation of cytotoxic activity.
