Simultaneous Determination of Urinary Androstanediol and Testosterone as an Evaluation of Human Androgenicity

Abstract

The amounts of α-androstane-3α, 17β-diol (androstanediol) and testosterone glucuronide excreted in the urine of 17 normal men, 10 normal women, 4 patients with testicular feminization syndrome, 10 women with idiopathic hirsutism and 10 women with acne vulgaris, were measured, following (β-glucuronidase hydrolysis, by gas-liquid chromatography. The mean (±SE) value for androstanediol in the urine of normal men was 133 ± 25 µg/24 hr and the corresponding mean value for testosterone was 59 ± 16 μg/24 hr. In normal women, the mean values for urinary androstanediol and testosterone were respectively 17 ± 6 μg/24 hr and 6.0 ± 2 μg/24 hr. In the 4 patients with the testicular feminization syndrome the mean urinary value of androstanediol (81 μg/24 hr) was close to that of testosterone (77 μg/24 hr) which is consistent with our previous in vivo findings suggesting that in these patients there is no formation of androstanediol outside the liver.
