Tissue Typing Using a Routine One‐Step Lymphocyte Separation Procedure

Abstract

S ummary . A reliable rapid one‐step method is described for the preparation of lymphocytes from 0.25 ml aliquots of defibrinated whole venous blood for tissue typing. An average yield of 70% lymphocytes is obtained relatively free from platelets, erythrocytes and granulocytes even in the presence of large numbers of granulocytes in blood obtained from cadaver donors. Using a high‐speed micro‐centrifuge lymphocytes can be prepared from defibrinated blood within 10 min and it is thus possible to complete a tissue typing test within 75 min. Using this method sufficient lymphocytes for typing purposes can be obtained from 0.5 ml of capillary blood thus greatly facilitating family studies where infants are involved.
