SUMMARY The addition of a goat antihuman γ-globulin reagent to the two-stage cytotoxicity test has produced a cell typing method which appears to be more sensitive than other widely-used variants of the lymphocytotoxicity technique. Twenty-six percent of 788 reactions negative with four conventional variants of cytotoxicity testing were positive with the antiglobulin method. The presence of antigen-antibody activity was confirmed by platelet absorption and elution studies for a number of these cytotoxicity negative-antiglobulin positive reaction combinations. The absorbed serum showed no cytotoxic activity against lymphocytes and the eluate recovered from the platelets gave positive tests by the antiglobulin method.
