A procedure for the purification of NAD kinase from pigeon liver acetone powder is described. Although not homogeneous, the preparation is largely free of enzymes which interfere with the assay of NAD kinase. The kinetics with magnesium as the activating ion are described: from Interrelationships between the kinetic coefficients, the modes of inhibition by NADH, NADPH and ADP, and the independent variation of V max and two K m s with pH, it is inferred that NAD kinase acts by a random addition, rapid equilibrium mechanism.
