A comparison of the Nuclear Overhauser Effect (NOE) between α‐carbon protons and amide protons in a protected dipeptide, the corresponding protected thionodipeptide and the corresponding cyclothionodipeptide shows that the acyclic thionodipeptide has the trans ‐configuration.
