Abstract

1. Callus tissue from the embryo of Secale cereale has been obtained and successfully maintained. Optimum conditions for callus initiation were achieved with dissected embryos transplanted to modified Heller's medium containing 2% sucrose, 0.5% yeast extract, and 1 mg/l of 2,4-D. 2. Subcultured callus tissue grew most successfully on this medium when it also contained casein hydrolysate. The maximum increase in fresh weight obtained in 25 days was 143.5%.
