Summary A brief survey of biochemical and pharmacological information on the action of the 4-amino folic acid antagonists is presented. An attempt is made to organize most of these facts into a coherent description of the mechanism of action of these drugs. It is concluded that, although their biochemical action is clearly a result of inhibition of the enzyme folic reductase, their chemotherapeutic value stems from differences in the permeability of various tissues to the compounds.
