Abstract

Normal 0 false false false IN X-NONE X-NONE MicrosoftInternetExplorer4 The endophytic fungi Chrisonilia sitophila , Acremonium sp ., and Penicillium sp . have been isolated from the tissues of the twigs of kandis gajah. All of the fungi strain s were grown in 3 L potato e dext r ose broth medium (PDB) at a room temperature for 28 days. To extract the antioxidant compounds, t he culture broth were filtered for mycelia removal followed by extraction and evaporation. All of the extracts were evaluated for their antioxidant activit ies by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. The extract of Acremonium sp. have strong activity with IC 50 value of 10.3 μg/mL, which is equivalent to ascorbic acid activity with IC 50 value of 9. 8 μg/mL. The extract was subjected to column chromatography on Si gel twice to obtain a high purity antioxidant compound in the form of yellow oil. The molecular structure was determined based on spectroscopic data, including 1 H-NMR, 13 C-NMR, HMQC, HMBC, and COSY. The compound was determined as sesquiterpene 3,5-di hydroxy -2,5-dimet hyl tride c a-2,9,11-trien e -4,8-dion e. /* Style Definitions */ table.MsoNormalTable {mso-style-name:"Table Normal"; mso-tstyle-rowband-size:0; mso-tstyle-colband-size:0; mso-style-noshow:yes; mso-style-priority:99; mso-style-qformat:yes; mso-style-parent:""; mso-padding-alt:0cm 5.4pt 0cm 5.4pt; mso-para-margin:0cm; mso-para-margin-bottom:.0001pt; mso-pagination:widow-orphan; font-size:11.0pt; font-family:"Calibri","sans-serif"; mso-ascii-font-family:Calibri; mso-ascii-theme-font:minor-latin; mso-fareast-font-family:"Times New Roman"; mso-fareast-theme-font:minor-fareast; mso-hansi-font-family:Calibri; mso-hansi-theme-font:minor-latin; mso-bidi-font-family:"Times New Roman"; mso-bidi-theme-font:minor-bidi;} Keywords: Acremonium sp., antioxidant compound, endophytic fungi , Garcinia griffithii , sesquiterpene /* Style Definitions */ table.MsoNormalTable {mso-style-name:"Table Normal"; mso-tstyle-rowband-size:0; mso-tstyle-colband-size:0; mso-style-noshow:yes; mso-style-priority:99; mso-style-qformat:yes; mso-style-parent:""; mso-padding-alt:0cm 5.4pt 0cm 5.4pt; mso-para-margin:0cm; mso-para-margin-bottom:.0001pt; mso-pagination:widow-orphan; font-size:11.0pt; font-family:"Calibri","sans-serif"; mso-ascii-font-family:Calibri; mso-ascii-theme-font:minor-latin; mso-fareast-font-family:"Times New Roman"; mso-fareast-theme-font:minor-fareast; mso-hansi-font-family:Calibri; mso-hansi-theme-font:minor-latin; mso-bidi-font-family:"Times New Roman"; mso-bidi-theme-font:minor-bidi;}
