The positions of the disulfide linkages of the kallikrein inactivator have been established. Disulfide bonds link half-cystine residues 5 and 55, 14 and 38, and 30 and 51. Reduction of these disulfide linkages yielded a completely inactive molecule. By slow oxidation of the reduced kallikrein inactivator, activity could be nearly completely restored.
